Anti-IL17A Antibody-Cytokine Fusion Protein, IgG (Vunakizumab)-IL22 (CAT#: VS-0624-FY84)
Vunakizumab is a recombinant humanized IgG, κappa monoclonal antibody that binds to and neutralizes the proinflammatory cytokine interleukin 17A (IL-17A). IL-22 is produced by several immune cell populations at sites of inflammation. IL-22 acts on non-hematopoietic cells, primarily stromal and epithelial cells. Its effects include stimulation of cell survival, proliferation, and synthesis of antimicrobial agents, including S100, Reg3β, Reg3γ, and defensins. This product is a novel bifunctional fusion protein, Vunakizumab-IL22 (vmab-mIL22), which has the activities of IL-22 and anti-IL-17A antibody. It can be used as a new approach to treat influenza A virus infection. vmab-mIL22 was constructed by genetically fusing mouse IL22 (mIL22) to the C-terminus of the heavy chain of anti-IL-17A antibody (Vunakizumab, vmab) via a (Gly4Ser)3 linker.

- IL-22 is an alpha-helical cytokine. IL-22 binds to a heterodimeric cell surface receptor composed of IL-10R2 and IL-22R1 subunits. IL-22R is expressed on tissue cells and absent on immune cells.
Crystallization was possible if the N-linked glycosylation site was removed in an IL-22 mutant that binds to the high-affinity cell surface receptor sIL-22R1. The crystal asymmetric unit contains two IL-22-sIL-22R1 complexes.

- The protein encoded by this gene is a proinflammatory cytokine produced by activated T cells. This cytokine regulates the activities of NF-kappaB and mitogen-activated protein kinases. This cytokine can stimulate the expression of IL6 and cyclooxygenase-2 (PTGS2/COX-2), as well as enhance the production of nitric oxide (NO). High levels of this cytokine are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis.
