The anti-IL17A pH-dependent antibody (VS-0825-YC175) targets human IL17A in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL17A acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL17A clearance. The antibody operates in a cyclical manner, binding IL17A in the extracellular fluid and releasing it in acidified endosomes. Human IL17A, or Interleukin-17A, is a pro-inflammatory cytokine that is a key driver of the Th17 immune response. It is involved in the immune response against extracellular pathogens and is implicated in various autoimmune diseases.
