The anti-IL23A pH-dependent antibody (VS-0825-YC188) targets human IL23 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL23 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL23 clearance. The antibody operates in a cyclical manner, binding IL23 in the extracellular fluid and releasing it in acidified endosomes. Human IL23, or Interleukin-23, is a cytokine that is a key driver of the Th17 immune response. It is involved in the pathogenesis of various autoimmune diseases, including psoriasis and inflammatory bowel disease.
