The anti-LAG3 pH-dependent antibody (VS-0825-YC236) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human LAG3. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for LAG3 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from LAG3 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human LAG3 (Lymphocyte-activation gene 3), also known as CD223, is a co-inhibitory receptor on T-cells that acts as an immune checkpoint molecule. It binds to MHC class II molecules to downregulate T-cell responses and is a target for cancer immunotherapy.
