Target

Introduction
Lymphocyte-activation gene 3 (LAG3) is an immune checkpoint receptor expressed on activated T cells, Tregs, B cells, and natural killer (NK) cells. It binds major histocompatibility complex class II (MHC-II) molecules and other ligands (e.g., FGL1), suppressing T cell proliferation, cytokine production, and cytotoxicity. Tumors exploit LAG3 to evade anti-tumor immunity, with overexpression linked to poor prognosis in melanoma, NSCLC, and breast cancer. LAG3 inhibitors (relatlimab) are approved in combination with PD-1 blockers for melanoma, leveraging synergistic immune activation. Ongoing trials test LAG3 targeting in other cancers, positioning it as a key next-generation checkpoint target.
