The anti-SNCA pH-dependent antibody (VS-0825-YC313) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to both human and mouse SNCA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for SNCA under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from SNCA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human SNCA, which encodes the alpha-synuclein protein, is a neuronal protein whose aggregation into structures known as Lewy bodies is a pathological hallmark of Parkinson's disease and other neurodegenerative disorders.
