The anti-TNFRSF10B pH-dependent antibody (VS-0825-YC334) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TRAIL-R2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFRSF10B under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFRSF10B at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFRSF10B, also known as TRAIL-R2 or DR5, is a death receptor that, like TRAIL-R1, binds to the TRAIL ligand to induce apoptosis. Its ability to trigger programmed cell death makes it another important therapeutic target in oncology.
