The anti-TNFRSF17 pH-dependent antibody (VS-0825-YC337) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TNFRSF17. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFRSF17 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFRSF17 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFRSF17, also known as BCMA, is the receptor for the cytokines BAFF and APRIL. It is essential for the survival of plasma cells, making it a key therapeutic target for the treatment of multiple myeloma.
