The Anti-TSHR Antibody Prodrug, Protease Activated (4G11) is an antibody prodrug comprised of a recombinant mouse antibody targeting human TSHR, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to TSHR protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to TSHR effectively. TSHR (Thyroid Stimulating Hormone Receptor) is a receptor whose signaling drives the growth and function of the thyroid gland. While not an oncogene itself, its signaling is often exploited in well-differentiated thyroid cancers. Tumors expressing TSHR can be treated by administering high doses of TSH (via TSH-stimulating hormones) to promote radioactive iodine uptake into the cancer cells.

Introduction
The protein encoded by this gene is a membrane protein and a major controller of thyroid cell metabolism. The encoded protein is a receptor for thyrothropin and thyrostimulin, and its activity is mediated by adenylate cyclase. Defects in this gene are a cause of several types of hyperthyroidism. Three transcript variants encoding different isoforms have been found for this gene.
