Recombinant Human Antibody (M22) is capable of binding to TSHR, expressed in HEK 293 cells. Expressed as the combination of a heavy chain (HC) containing VH from anti-TSHR mAb and CH1-3 region of human IgG and a light chain (LC) encoding VL from anti- TSHR proteins mAb and CL of human kappa light chain. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition. A human monoclonal autoantibody to the TSHR (M22), which is a powerful thyroid stimulator with high affinity for the receptor, and determined its crystal structure at 1.65A° resolution.
Figure 1 TSHR-stimulating, but not blocking, mAbs to the TSHR induce cAMP production in human thymocytes.
Intracellular cAMP was measured by ELISA after 1-h incubation with the indicated amounts of the mAb with stimulating activity M22 (dotted line), the monoclonal thyroid-blocking Ab K1-70 (solid line), or a normal mouse IgG (dashed line) in human thymocytes. Bars represent mean ± SEM of three to four experiments. All experiments are performed in triplicates; *p < 0.05.

Figure 2 Kinetics of ¹²⁵I-labeled TSH, M22 IgG, and M22 Fab interacting with TSHR coated tubes.
(A)Time course of binding of ¹²⁵I-labeled TSH, M22 IgG, and M22 Fab (nonspecific binding, i.e., binding of labeled TSH, M22IgG, or M22 Fab to tubes not coated with the TSHR was low (2–2.6% of total cpm added) and was not subtracted from thedata shown in the figure).
