Recombinant Mouse Antibody (79D) is capable of binding to Kit. The inhibitory KIT antibodies that bind to the membrane-proximal Ig-like D4 of KIT with significant overlap with an epitope in D4 that mediates homotypic interactions essential for KIT activation.

NIH 3T3 cells expressing WT KIT were incubated for 5 h with the indicated concentrations (Upper) of Fab79D following stimulation with 25 ng/mL of SCF for 5 minat 37 °C. Lysates of unstimulated or SCF-stimulated cells were subjected to immunoprecipitation (IP) with anti-KIT antibodies followed by SDS/PAGE and immunoblotting (IB) with anti-KIT or anti-phosphotyrosine (pY) anti-bodies.
