The anti-BRAF pH-dependent antibody (VS-0825-YC220) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human BRAF. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for BRAF under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from BRAF at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human BRAF (B-Raf proto-oncogene, serine/threonine kinase) is a key component of the MAPK/ERK signaling pathway. Oncogenic mutations in the BRAF gene, most notably the V600E mutation, are a primary driver of several cancers, including melanoma and thyroid cancer.
