SmartAb™ Recombinant Anti-CA9 pH-dependent Antibody (Clone Girentuximab)

The anti-CA9 pH-dependent antibody (clone Girentuximab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human carbonic anhydrase IX (CAIX). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CAIX under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CAIX at pH 6.0. This pH-sensitive antibody retains its binding affinity for CAIX on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human Carbonic Anhydrase IX (CAIX) is a transmembrane enzyme that helps regulate pH in the tumor microenvironment. It is overexpressed in many types of solid tumors, particularly under hypoxic conditions, and is associated with a poor prognosis.
