The anti-DLL4 pH-dependent antibody (VS-0825-YC102) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human DLL4. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for DLL4 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from DLL4 at pH 6.0. This pH-sensitive antibody retains its binding affinity for DLL4 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human DLL4, or Delta-like protein 4, is a transmembrane protein that is a key ligand for the Notch signaling pathway. It is crucial for vascular development and angiogenesis, and its signaling is often targeted to inhibit tumor growth.
