The anti-IL1B pH-dependent antibody (VS-0825-YC182) targets human IL1-beta in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL1beta acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL1-beta clearance. The antibody operates in a cyclical manner, binding IL1-beta in the extracellular fluid and releasing it in acidified endosomes. Human IL1-beta, or Interleukin-1 beta, is a potent pro-inflammatory cytokine that is a central mediator of inflammation and fever. It is a key component of the innate immune response and is implicated in a wide range of inflammatory and autoimmune diseases.
