The anti-IL2 pH-dependent antibody (VS-0825-YC184) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for IL2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IL2, or Interleukin-2, is a cytokine that is crucial for the proliferation, differentiation, and survival of T-cells. It is a central player in the adaptive immune response and is used clinically as an immunotherapy for certain cancers.
