The anti-IL20 pH-dependent antibody (clone Fletikumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IL20. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL20 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL20 at pH 6.0. This pH-sensitive antibody retains its binding affinity for IL20 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IL20, or Interleukin-20, is a cytokine that is involved in skin inflammation and is a key player in the pathogenesis of psoriasis. It is a member of the IL-10 family and its signaling promotes the proliferation of keratinocytes.
