- Introduction
- MET is a transmembrane tyrosine kinase receptor binding hepatocyte growth factor (HGF, scatter factor). Expressed on epithelial cells, it regulates cell proliferation, migration, invasion, and angiogenesis-critical for tissue repair and development. Aberrations (amplification, mutation, overexpression) drive tumorigenesis in NSCLC, gastric cancer, hepatocellular carcinoma, and papillary renal cell carcinoma. MET amplification is a common resistance mechanism to EGFR TKIs in NSCLC. Targeted therapies include TKIs (capmatinib, tepotinib) and monoclonal antibodies, blocking HGF-MET signaling to inhibit tumor growth and metastasis. MET targeting addresses both primary tumors and EGFR TKI-resistant cancers.
