SmartAb™ Recombinant Anti-TEK pH-dependent Antibody (Clone 15B8)

The anti-TEK pH-dependent antibody (clone 15B8) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TEK. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TEK under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TEK at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TEK (Tyrosine Kinase with Immunoglobulin-like and EGF-like domains), also known as Tie2, is a receptor tyrosine kinase that binds to angiopoietin ligands. It is a key regulator of angiogenesis and vascular stability.
