The anti-TNFSF11 pH-dependent antibody (VS-0825-YC343) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TNFSF11. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFSF11 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFSF11 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFSF11, also known as RANKL, is a cytokine of the TNF family. It is essential for the formation and activation of osteoclasts and is a key regulator of bone remodeling. Therapies targeting TNFSF11 are used to treat bone disorders such as osteoporosis.
