The anti-TNFSF13B pH-dependent antibody (VS-0825-YC344) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human BLyS. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFSF13B under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFSF13B at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFSF13B, also known as BLyS or BAFF, is a cytokine of the TNF family that is essential for the survival and maturation of B-cells. Its dysregulation is implicated in autoimmune diseases such as lupus.
