    This research investigates the development of engineered H-Ferritin (HFn) nanocages for targeted delivery of navitoclax (Nav) to cancer-associated fibroblasts (CAFs) in the tumor microenvironment. The study demonstrates that by functionalizing HFn nanocages with fibroblast activation protein (FAP) antibody fragments, they achieved significantly higher binding with FAP-positive CAFs compared to FAP-negative cancer cells. The navitoclax-loaded nanocages (HNav-FAP) showed enhanced cytotoxicity specifically in FAP-positive cells while maintaining the drug's pro-apoptotic mechanism of action. Importantly, in vivo experiments confirmed that HFn-FAP successfully reached tumors and targeted CAFs after intravenous administration in a mouse model of triple-negative breast cancer.
    Creative Biolabs provided FAP antibody fragments (CAT#: TAB-024WM-F(E)) that were used to functionalize the HFn nanocages. These antibody fragments enabled the selective targeting of CAFs over cancer cells by recognizing FAP, a cell surface glycoprotein highly expressed in the stroma surrounding epithelial cancers but not in healthy tissues. This targeting capability was fundamental to the study's success in developing a promising therapeutic approach that could enhance specific drug delivery into CAFs while overcoming navitoclax's limitations of hydrophobicity and toxicity, potentially opening new avenues for tumor microenvironment-focused cancer therapy.
