Anti-TNFRSF9 immunotoxin (scFv)-Sap (CAT#: AGTO-G063S)
This immunotoxin was achieved by conjugating/fusing the Anti-TNFRSF9 scFv to saporin. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to the cell surface TNFRSF9 and enters cells by receptor-mediated endocytosis. Upon release of the conjugate from endosomes, the saporin toxin catalytically renders the large subunit ribosomes unable to bind the elongation factors and inhibits protein synthesis. It was designed for treatment of graft-versus-host diseas (GVHD) and the relapse of leukemia.

- Saporin is a so-called ribosome inactivating protein (RIP), due to its N-glycosidase activity, from the seeds of Saponaria officinalis (common name: soapwort).Saporin depurinate the ribosomal RNA in a specific position; rendering the large subunit of ribosomes unable to bind the elongation factors. The consequent arrest of protein synthesis is responsible for the cytotoxicity.
