    The anti-CD2 pH-dependent antibody (VS-0825-YC39) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD2, also known as LFA-2, is a cell adhesion molecule found on the surface of T-cells and NK cells. It binds to CD58 on antigen-presenting cells to promote cell-cell adhesion and co-stimulate T-cell activation.
