The anti-CD276 pH-dependent antibody (VS-0825-YC48) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD276. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD276 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD276 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD276 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD276, also known as B7-H3, is a transmembrane protein belonging to the B7 family of co-stimulatory molecules. It is expressed on a variety of cells, and its exact function is still being investigated, but it is known to play a role in both promoting and inhibiting T-cell responses.
