The anti-CD40LG pH-dependent antibody (VS-0825-YC58) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD40L. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD40L under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD40L at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD40L on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD40L, also known as CD154, is a protein expressed on activated T helper cells that binds to CD40 on B-cells and other antigen-presenting cells. This interaction is essential for the activation of these cells and plays a critical role in adaptive immunity.
