The anti-CEACAM5 pH-dependent antibody (clone Altumomab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CEACAM5. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CEACAM5 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CEACAM5 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CEACAM5 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CEACAM5, also known as CEA, is a cell surface glycoprotein that is a well-known tumor marker for various cancers, particularly colorectal cancer. It is involved in cell adhesion and is a target for diagnostic and therapeutic antibodies.
