The anti-FGFR2 pH-dependent antibody (VS-0825-YC132) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human FGFR2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for FGFR2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from FGFR2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for FGFR2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human FGFR2, or Fibroblast Growth Factor Receptor 2, is a receptor tyrosine kinase that is involved in embryonic development, particularly in bone formation and limb development. Mutations in its gene are linked to skeletal disorders and its overexpression is a key driver of certain cancers.
