SmartAb™ Recombinant Anti-NOTCH1 pH-dependent Antibody (Clone Brontictuzumab)

The anti-NOTCH1 pH-dependent antibody (clone Brontictuzumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human NOTCH1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for NOTCH1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from NOTCH1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human NOTCH1 is a transmembrane receptor that is a key component of the Notch signaling pathway, which regulates cell fate decisions. Its dysregulation is linked to various developmental disorders and cancers, most notably T-cell acute lymphoblastic leukemia (T-ALL).
