Anti-TGFBR2 Antibody Prodrug, Protease Activated (TGF1)

The Anti-TGFBR2 Antibody Prodrug, Protease Activated (TGF1) is an antibody prodrug comprised of a recombinant human antibody targeting human TGFBR2, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to TGFBR2 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to TGFBR2 effectively. TGFBR2 (Transforming Growth Factor Beta Receptor 2) is a key component of the receptor complex that mediates signaling for the tumor suppressor cytokine TGF-beta. Inactivating mutations in TGFBR2 are common in cancers, particularly those with microsatellite instability (MSI), such as colorectal cancer. Loss of this receptor prevents the tumor cell from receiving the growth-inhibitory signals of TGF-beta, thus enabling uncontrolled proliferation.

- This gene encodes a member of the Ser/Thr protein kinase family and the TGFB receptor subfamily. The encoded protein is a transmembrane protein that has a protein kinase domain, forms a heterodimeric complex with another receptor protein, and binds TGF-beta. This receptor/ligand complex phosphorylates proteins, which then enter the nucleus and regulate the transcription of a subset of genes related to cell proliferation. Mutations in this gene have been associated with Marfan Syndrome, Loeys-Deitz Aortic Aneurysm Syndrome, and the development of various types of tumors. Alternatively spliced transcript variants encoding different isoforms have been characterized.
