SmartAb™ Recombinant Anti-CCL2 pH-dependent Antibody (Clone C775)

The anti-CCL2 pH-dependent antibody (clone C775) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CCL2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CCL2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CCL2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CCL2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CCL2, also known as Monocyte Chemoattractant Protein-1 (MCP-1), is a chemokine that recruits monocytes, macrophages, and other immune cells to sites of inflammation. It is a major driver of chronic inflammation and is implicated in various diseases, including atherosclerosis and cancer.
