SmartAb™ Recombinant Anti-CD86 pH-dependent Antibody (Clone Hu3D1)

The anti-CD86 pH-dependent antibody (clone Hu3D1) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human B7-2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD86 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD86 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD86 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human B7-2, also known as CD86, is a co-stimulatory molecule expressed on antigen-presenting cells. It provides the second signal for T-cell activation by binding to CD28 and CTLA-4, and its interaction with CD28 is crucial for the initiation of the adaptive immune response.
