anti-CD86 immunotoxin 1G10 (IgG)-Bouganin (CAT#: AGTO-L002O)

This immunotoxin was achieved by conjugating/fusing the anti-CD86 monoclonal antibody to Bouganin. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to CD86-expressing cells. Once bind, the toxin enters cells by receptor-mediated endocytosis and enzymatically damages ribosomes, ultimately leading to cell death. It was designed for treatment of Hodgkin's disease or other CD86-expressing tumours.

  • Bouganin is a type-I ribosome inactivating protein (RIP) that was recently purified from leaves of Bougainvillea spectabilis Willd. Like all type-I RIPs, bouganin is a RNA glycosidase with deadenylating activity. Bouganin was characterized as a single-chain protein with a molecular mass of 26 200 Da and its mature protein consists of 250 amino acids. Furthermore, it was found that bouganin is probably the least toxic RIP known based on protein inhibiting studies in cell lines. It was observed that the bouganin concentrations inhibiting protein synthesis of human cell lines were much higher than the concentrations required to inhibit protein synthesis in a rabbit reticulocyte lysate assay, a characteristic common to all type-I RIPs.
