The anti-TGFB1 pH-dependent antibody (VS-0825-YC326) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TGFB1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TGFB1 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TGFB1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TGFB1 (Transforming Growth Factor Beta 1) is a specific isoform of the TGF-β cytokine family. It is a key regulator of immune responses, wound healing, and tissue fibrosis.
