The anti-CD37 pH-dependent antibody (clone Lilotomab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD37. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD37 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD37 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD37 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD37 is a transmembrane protein of the tetraspanin family expressed on B-cells. It is involved in B-cell signaling and adhesion, making it a target for antibody-drug conjugates in the treatment of B-cell lymphomas.
