The anti-CD44 pH-dependent antibody (clone Bivatuzumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD44. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD44 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD44 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD44 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD44 is a cell surface adhesion molecule involved in cell-cell and cell-matrix interactions. It serves as a receptor for hyaluronic acid and is involved in cell migration, proliferation, and the progression of various cancers.
