The anti-PDCD1 pH-dependent antibody (VS-0825-YC19) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human PDCD1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for PDCD1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from PDCD1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for PDCD1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human Programmed Cell Death Protein 1 (PDCD1) is a co-inhibitory receptor expressed on activated T-cells that helps regulate the immune response. When it binds to its ligands (PD-L1 and PD-L2), it delivers an inhibitory signal that prevents T-cells from attacking other cells, a pathway often exploited by cancer to evade immune surveillance.
