The antibody binds to human PD-1 with pH-dependent antigen binding properties. The antibody preferred PDCD1 binding under the acidic pH relative to physiological pH by introducing His mutations, which could act as a pH-switch. By employing yeast surface display with a pH-dependent screening pressure, mAbs having markedly increased dissociation from PDCD1 at pH 6.0 were identified. The pH-sensitive antibody maintained a binding affinity on tumor cells, while having negligible binding to PDCD1 expressed on normal cells under physiological pH. Human PD-1, or Programmed Cell Death Protein 1, is a co-inhibitory receptor expressed on activated T-cells that helps regulate the immune response. When it binds to its ligands (PD-L1 and PD-L2), it delivers an inhibitory signal that prevents T-cells from attacking the cell.
