This ADC product is composed of an anti-CD70 antibody (MDX-1115) conjuagated to Duocarmycins (MDX-1115-Duocarmycins). It has demonstrated a response in NHL treatment by a MOA (Mechanism of Action) of Bind to DNA minor groove and cause DNA damage.

- The protein encoded by this gene is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for TNFRSF27/CD27. It is a surface antigen on activated, but not on resting, T and B lymphocytes. It induces proliferation of costimulated T cells, enhances the generation of cytolytic T cells, and contributes to T cell activation. This cytokine is also reported to play a role in regulating B-cell activation, cytotoxic function of natural killer cells, and immunoglobulin sythesis.

High Potency CD70 Targeting
This ADC targets CD70 (TNFSF7) effectively. We observed that the MDX-1115 antibody bound to activated lymphocytes. The Duocarmycin payload is an extremely potent DNA alkylating agent, binding to the minor groove and causing significant DNA damage.
Efficient Di-Peptide Cleavage
The cleavable di-peptide linker used in this conjugate proved to be stable in our media but was rapidly cleaved by intracellular proteases. This released the toxin specifically within the target cells, allowing us to study the effects of DNA damage in NHL models.
