This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EpCAM scFv-CH1; scFv-Cκ to IL2 and GM-CSF. It was expressed in CHO and purified with affinity chromatography. The Immunocytokines retains the ability to bind the EpCAM as well as the biological activity of IL2 and GM-CSF. The bi-antibody-cytokine fusion protein exhibited the ability to bind the targeted antigen and retained the biological activity of murine IL2 and GM-CSF to induce CTLL-2 and FDC-P1 cell proliferation respectively. DCH induced human PBMC and T-cell-mediated cytotoxicity of Kato III cells (from a human gastric carcinoma that over-express the EpCAM antigen). This Immunocytokines was designed for treating EpCAM over-expressing cancers.
