The Anti-CD274 Antibody Prodrug, Protease Activated (VS-1025-YC39) is an antibody prodrug comprised of a recombinant human antibody targeting human CD274, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to CD274 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to CD274 effectively. CD274, or PD-L1, is an immune checkpoint molecule frequently expressed by tumor cells to achieve immune evasion. By binding to the PD-1 receptor on T-cells, it delivers an inhibitory signal, effectively stopping the T-cell from recognizing and killing the cancer. Blockade of the PD-1/PD-L1 axis with antibodies is a cornerstone of modern immunotherapy.

Target

Introduction
This gene encodes an immune inhibitory receptor ligand that is expressed by hematopoietic and non-hematopoietic cells, such as T cells and B cells and various types of tumor cells. The encoded protein is a type I transmembrane protein that has immunoglobulin V-like and C-like domains. Interaction of this ligand with its receptor inhibits T-cell activation and cytokine production. During infection or inflammation of normal tissue, this interaction is important for preventing autoimmunity by maintaining homeostasis of the immune response. In tumor microenvironments, this interaction provides an immune escape for tumor cells through cytotoxic T-cell inactivation. Expression of this gene in tumor cells is considered to be prognostic in many types of human malignancies, including colon cancer and renal cell carcinoma. Alternative splicing results in multiple transcript variants.
