    The anti-CD80 pH-dependent antibody (clone Galiximab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD80. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD80 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD80 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD80 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD80, also known as B7-1, is a co-stimulatory molecule expressed on antigen-presenting cells. It binds to both CD28 and CTLA-4 on T-cells, providing the second signal required for T-cell activation and playing a critical role in regulating the immune response.
