This product is a recombinant mouse antibody that recognizes MUC1 (Mucin1). The original antibody was secreted by the hybridoma and the recombinant antibody was expressed in mammalian cells with chemically defined culture media and was purified by affinity chromatography. This antibody is against the immunogenic glycopeptide of MUC1.

Dot Blot analysis of TAB-418MZ was performed by coating with human MUC1 protein (His tag).
TAB-418MZ incubation concentration: 2 μg/mL.
HRP-conjugated goat anti-mouse IgG was used as a secondary antibody (1: 2000)

Western blot analysis of TAB-418MZ was performed with human MUC1 protein onto a 12% Tris-HCl polyacrylamide gel. Proteins were transferred to a CN membrane and blocked with 5% skim milk for at least one hour. Membranes were probed with TAB-418MZ and HRP goat anti-mouse IgG was used as a secondary antibody (1: 2000). Chemiluminescent detection was performed.
Lane 1: Reduced antigen (0.1 μg)
Lane 2: Reduced antigen (0.3 μg)
Lane 3: Reduced antigen (0.6 μg)

SDS-PAGE analysis of TAB-418MZ in non-reduced (Lane 1) and reduced (Lane 2) conditions. Gel stained for 30 minutes with Coomassie Blue. As a result, different β-mercaptoethanol-reduced proteins (Heavy chain and Light chain) migrate as about 50 kDa and 25 kDa, respectively.
The purity of TAB-418MZ was greater than 95% as determined by SDS-PAGE

The study investigated the interaction between colon cells and macrophages in the context of ulcerative colitis (UC) and colitis-associated colon cancer (CACC). Researchers found that macrophages, particularly the M2-like phenotype, play a significant role in promoting the expression of the glycosyltransferase ST6GALNAC1 in colon cancer cells. This enzyme catalyzes the addition of sialic acid to O-linked GalNAc residues, leading to the formation of tumor-associated sialyl-Tn (sTn) antigens on the mucin MUC1. The study demonstrated that the presence of M2 macrophages, through cytokines such as IL13 and CCL17, increased the expression of ST6GALNAC1 and the corresponding MUC1-sTn glycoform, contributing to the pathogenesis and progression of UC and CACC.
