  • 1. Reduction of Antibody Fragments:
    Reduce W24HD37-CCH using 5 mM DTT in HBS (pH 8.0) for 15-20 minutes at room temperature.
    2. Removal of Unreacted Reducing Agent:
    Remove unreacted DTT by chromatographic separation using a Sephadex G-25 column equilibrated with degassed HBS (pH 7.4).
    3. Coupling to Liposome Mediators:
    Combine W24HD37-CCH with Mal-PEG2000-DSPE particles at a molar ratio of 1:4 (antibody fragment: Mal-PEG2000-DSPE) and stir at room temperature for 16 hours.
    4. Thiolating Antibodies:
    Treat W24HD37 monoclonal antibody (mAb) with Traut's reagent at a molar ratio of 10:1 (Traut's reagent: W24HD37 mAb) for 1 hour using degassed HBS (pH 8.0).
    5. Removal of Unreacted Traut's Reagent:
    Remove unreacted Traut's reagent by chromatographic separation using a Sephadex G-50 column equilibrated with degassed HBS (pH 7.4).
    6. Coupling to Liposome Mediators:
    Combine thiolated W24HD37 mAb with a mixture of Mal-PEG2000-DSPE/mPEG2000-DSPE at a molar ratio of 1:10 (mAb: Mal-PEG2000-DSPE) and stir at room temperature for 16 hours.
    7. Liposome Formation and Antibody Insertion:
    Incubate the particles bound to Mal-PEG2000-DSPE (W24HD37-CCH, or W24HD37 mAb) with pre-formed liposomes (SIL) for 16 hours at room temperature with continuous stirring to facilitate the insertion of antibodies or fragments.
    8. Separation:
    Separate using a Sepharose CL-4B column under HBS (pH 7.4) conditions to remove unbound antibodies and free liposomes.
