FGK45 was employed in the formulation of anti-CD40 antibody-mediated bufalin liposomes, specifically designed to induce apoptosis in B16 melanoma cells. - Immunoliposomes Method
- To prepare bufalin liposomes conjugated with anti-CD40 antibodies, the researchers first coupled the anti-CD40 monoclonal antibody with functionalized liposomes through a reaction between maleimide and thiol groups. The specific method involved mixing the antibody with excess DTT and 10 mM EDTA, reacting at 25°C for 20 minutes, and then removing unreacted DTT using a dialysis column. Subsequently, the exposed thiol residues on the antibody were allowed to conjugate at a ratio of 1 mg of antibody to 2 mmol of liposomes at 25°C for at least 10 hours.
The resulting liposome suspension was then extruded five times using a LipoFast-Pneumatic Lipex extruder with a 0.2 μm pore size polycarbonate membrane to form unilamellar liposomes. After that, the preparation was subjected to centrifugation and multiple washes with PBS to remove unbound antibodies and bufalin.

- The FGK45 monoclonal antibody specifically targets mouse CD40, also referred to as Bp50. Its immunogen consists of a recombinant mouse CD40 fusion protein. The FGK45 antibody functions as an agonistic antibody, demonstrating the capability to activate antigen-presenting cells (APCs) that express CD40. Additionally, FGK45 can be employed to inhibit the CD40/CD154 interaction both in vitro and in vivo.
