The anti-CD40 pH-dependent antibody (VS-0825-YC57) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD40. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD40 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD40 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD40 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD40 is a co-stimulatory molecule expressed on antigen-presenting cells, including B-cells and macrophages. Its interaction with its ligand, CD40L, is crucial for B-cell activation, isotype switching, and T-cell-dependent immune responses.
