Anti-EpCAM Antibody-Cytokine Fusion Protein, IgG (KS-1/4)-IL12-IL2 (CAT#: ACFP-SH179)

This antibody-cytokine fusion protein was achieved by conjugating/fusing the Anti-EpCAM IgG to IL12 and IL2. It was expressed in CHO and purified with affinity chromatography. The Immunocytokines retains the ability to bind the EpCAM as well as the biological activity of IL12 and IL2. KS-IL12-IL2 caused a profound inhibition of tumor growth, the multiple-cytokine antibody fusion protein can be used to treat cancer in an immunosuppressed animal. In the context of bi-functional fusion proteins, hetero-dimeric IL-12 could be expressed either in a single-chain form, or maintained as a heterodimer in which the p40 subunit was fused to IL-2. These IL-12/IL-2 bi-functional fusion proteins were shown to induce extremely high levels of interferon-gamma (IFN-γ), similar to the synergy normally seen with the combined application of the individual cytokines.   • Interleukin 12 (IL-12) is an interleukin that is naturally produced by dendritic cells, macrophages, neutrophils, and human B-lymphoblastoid cells (NC-37) in response to antigenic stimulation. IL-12 is composed of a bundle of four alpha helices. It is a heterodimeric cytokine encoded by two separate genes, IL-12A (p35) and IL-12B (p40). The active heterodimer (referred to as 'p70'), and a homodimer of p40 are formed following protein synthesis.
