The anti-IL2RA pH-dependent antibody (clone MAK179) targets human IL2RA in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL2RA acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL2RA clearance. The antibody operates in a cyclical manner, binding IL2RA in the extracellular fluid and releasing it in acidified endosomes. Human IL-2RA, also known as CD25, is a subunit of the high-affinity IL-2 receptor that is expressed on activated T-cells and regulatory T-cells. It is essential for IL-2 signaling and plays a critical role in T-cell proliferation and survival.
