The Human ESR1 Promoter Primers offer a highly specific solution for amplifying DNA obtained through chromatin immunoprecipitation (ChIP). These optimized forward and reverse PCR primers are precisely designed to target a region of the human estrogen receptor 1 (ESR1) promoter. Upon ligand binding, the estrogen receptor forms dimers and binds to estrogen response elements on various genes crucial for cell growth and sexual development. Deregulation of ESR1 is implicated in breast and endometrial cancers. These primers are thoroughly optimized for SYBR® Green quantitative real-time PCR and validated with Enzymatic Chromatin IP Kits and ChIP-validated antibodies, facilitating research into estrogen signaling and related pathologies.

- The core function of primers in ChIP experiments is to serve as specific probes for detecting and quantifying particular DNA fragments enriched after immunoprecipitation, thereby revealing protein-DNA interactions.
- Workflow
- 1. Label PCR tubes/plates for duplicates, no-DNA control, and a 2% total input chromatin DNA serial dilution.
2. Add 2 μl ChIP DNA sample per well/tube.
3. Prepare and add 18 μl master PCR reaction mix (Nuclease-free H2O, Primers, SYBR® Green).
4. Run 40 cycles of PCR: 95°C for 3 min; then 95°C for 15 sec, primer-specific temp for 60 sec (repeated).
5. Analyze results with real-time PCR software.

- Introduction
- This gene encodes an estrogen receptor, a ligand-activated transcription factor composed of several domains important for hormone binding, DNA binding, and activation of transcription. The protein localizes to the nucleus where it may form a homodimer or a heterodimer with estrogen receptor 2. Estrogen and its receptors are essential for sexual development and reproductive function, but also play a role in other tissues such as bone. Estrogen receptors are also involved in pathological processes including breast cancer, endometrial cancer, and osteoporosis. Alternative promoter usage and alternative splicing result in dozens of transcript variants, but the full-length nature of many of these variants has not been determined.
