ARG2: A Mitochondrial Regulator of Vascular Health and Cell Growth
Arginase 2 (ARG2) is a mitochondrial enzyme that catalyzes the hydrolysis of L-arginine to L-ornithine and urea, similar to the cytosolic arginase 1 (ARG1) enzyme. Unlike ARG1, ARG2 is expressed in extrahepatic tissues and predominantly localized in the mitochondria. In endothelial cells, ARG2 modulates NO production by competing with nitric oxide synthase (NOS) for their common substrate L-arginine. The overexpression of ARG2 in endothelial cells has been linked to reduced NO bioavailability, leading to endothelial dysfunction and contributing to the pathogenesis of diseases like atherosclerosis and pulmonary hypertension. ARG2 also plays a role in polyamine biosynthesis, which is crucial for cell proliferation and growth, a property being exploited in cancer research.

Background
Arginase catalyzes the hydrolysis of arginine to ornithine and urea. At least two isoforms of mammalian arginase exists (types I and II) which differ in their tissue distribution, subcellular localization, immunologic crossreactivity and physiologic function. The type II isoform encoded by this gene, is located in the mitochondria and expressed in extra-hepatic tissues, especially kidney. The physiologic role of this isoform is poorly understood; it is thought to play a role in nitric oxide and polyamine metabolism. Transcript variants of the type II gene resulting from the use of alternative polyadenylation sites have been described.
